| OBJICTIVE:To investigate the reversal effect of MDR1 and MRP antisense oligodooxynucleotide(ASODN) on adriamycin-resistant SGC7901/ADM cells.METHODS: The antisense-phosphorothioate oligodeoxynucleotide which combined to the specific fragment of MDR1 and MRP gene mRNA was artificially synthesized, meanwhile, the sense oligodooxynucleotide (SODN) was synthesized as control. Lipofectamine 2000(LF) was used for the transfer of either ASODN or SODN into SGC7901/ADM cells. After transfected by MDR1 and MRP ASODN , the sensitivity of SGC7901/ADM cells to adriamycin and other anticancer drugs,the intracellular fluorescence intensity of Rhodamine 123 ,the change of cell cycle,the expression of MDR1 and MRP in gene and protein level were detected repectively by (MTT)assay,flow cytometry(FCM),RT-PCR and immunocytocytochemistry.RESULTS :1.
