| [Objective] The aim of this study is To evaluate the expression of mRNA and proteinon Kiss-1 and p27Kip1 gene in osteosarcoma cell MG-63 ,U2 and tissue, and to explorethe relationship between the level of expression and the biologic character ofostosarcoma, and to establish the foundation of allied gene therapy and fused genetherapy targeting at these two genes against osteosarcoma.[methods] 1.The osteosarcoma cell lines MG-63, U2 and osteoblast cell line OB arecultured.2.The expression level of mRNA of Kiss-1 and p27Kip1 gene is assessed in the three cell lines with the semiquantitative RT-PCR.3.The expression level of protein of Kiss-1 and p27Kip1 gene is evaluated immunocytochemically in the three cell lines.4.The expression level of protein of Kiss-1 and p27Kip1 gene is investigated immunohistochemically in the histological section of osteosarcoma, ostochondroma and normal bone tissue.[results] 1. The cell line MG-63 doesn't express the mRNA of Kiss-1 gene. The expression level of Kiss-1 mRNA in cell line OB is higher than which in cell line U2. The expression level of Kiss-1 protein in cell line OB is higher than which in cell line U2. The cell line MG-63 doesn't express the protein of Kiss-1 gene.2. There is no difference on the expression level of p27Kip1 mRNA between the three cell lines MG-63, U2 and OB. The expression level of p27Kip1 protein in cell line OB is higher than which in cell lines MG-63 and U2. There is no difference on the expression level of p27~(Kip1) protein between these two osteosarcoma cell lines. |
