| ObjectiveTo observe the expression of cyclin-dependent kinase-4(CDK4) cyclin-dependent kinase inhibitor(P16) and Hypoxia Inducible Factor 1 [alpha](HIF-1α) in human middle ear cholesteatoma and to explore their role in the development of cholesteatoma.MethodThe materials used in this study were 35 acquired cholesteatomas tissues collected from patients in the age 10-56 years during surgery,20 normal human external ear canal skin were used as controls. All specimens were stained immunohistochemical SABC method for CDK4,P16 and HIF-1α. Then specimens were analyzed under microscope respectively. Correlations between the indexes and relationship between indexes and the degree of ossicular destruction were investigated.Result1. CDK4, P16 were expressed in parabasal layers and the upper layer, mainly in the prickle and granular layers of cholesteatoma epithelium. Their positive rates were 68.6% and 88.6% respectively.
