| Objective : The injury and dysfunction of endothelial cells may be one of the important factor responsible for the pathogenesis of hypertensive disorder complicating pregnancy . The vascular endothelium produces a number of substances in which endothelial-derived relaxing factor or nitric oxide (NO) and endothelin (ET) are thought to be important . Now the causes and mechanisms are not clear yet , recently more and more findings suggest that there is a close correlation between preeclampsia , a kind of hypertensive disorder complicating pregnancy , and the imbalance between NO and ET . Low molecular weight heparin (LMWH) is widely used clinically , but it's non-coagulation functions are not known yet . To investigate the effects of LMWH on expression of endothelium nitric oxide synthase (eNOS) - messenger ribonucleic acid (mRNA) and ET and the percentages of different cell cycles , the human umbilical vein endothelial cells (HUVEC) - ECV304 were cultivated with the serum either from preeclampsia or from normal late trimester pregnant women , then stimulated by LMWH .Methods : HUVEC-ECV304 were incubated with medium contain 20% serum (volume/volume) either from preeclampsia (n=18) or from normal late trimester pregnant women (n=12) matched for maternal age , gestational age and parity . Blood samples were collected before the administration . HUVEC-ECV304 , were passaged to five 6-well dishes (with a piece of aseptic glass in each well) every time , then incubated with the same serum every three wells for 48 hours . The cells in the same serum were divided into three subgroups by random : the low and high concentration of LMWH groups (1U/ml or 2U/ml final concentration) and... |
