In this research, the acute toxicity test, microcleus test of born marrow, chromosome aberration test of bom marrow cell, sperm shape abnormality test, teratogenicity test and cumulative toxicity test were used. The main results of toxicological evaluation are the following:1. Acute toxicity test: The median lethal dose (LD50) peroral on mouse is 2100mg/kg.bw. 95% confidence interval is 18202531 mg/kg.bw. According to the standard of acute toxic classification, Sodium Benzoate is low-toxic whose toxicity is the third-class.2. Microcleus test of born marrow cell in mice: In positive control group (♀38.8±0.77‰, ♂40.6±0.719‰), high-dose group (♀25.5±0.71‰, ♂31.5±2.12‰) and mid-dose group (♀14.5±0.71‰, ♂20.4±0.71‰), there was significant difference between negative control group (1.8‰) (p<0.01). And there was dose-response relationship between them. So Sodium Benzoate is chromosome clastogen. And the mutagenesis to male is greater than to female.3. Chromosome aberration test of born marrow cell: In this test, the frequency of chromosomal aberration in three experimental groups were 39.00%, 28.75%, and 5.75%.There was marked difference in positive control group (45.75%) compared with negative control group (1.13%) (p<0.01). In mid-, high-dose groups, the frequency of chromosomal aberration of mice was significantly higher than that in the negative control group (p<0.01). And there was dose-response relationship between them. So Sodium Benzoate has the function of chromosome clastogen. And isochromatid break is the main type of chromosome aberration.4. Sperm shape abnormality test: In this test, the frequency of Sperm abnormalities induced by Sodium Benzoate in mice in three experimental groups were 6.3%, 5.5%, and 5.3%.Though statistic analysis, there was marked difference in positive control group (7.1%) and other three dose groups compared with negative control group (2.1%). So Sodium Benzoate can result in genetic toxicity. Within the sperm abnormalities, the amount of bend tail is the most.5. Teratogenicity test: Compared with negative control group, there was no marked difference about weight and length of offspring. Three experimental groups all didn't have malformation in the embryo appearance, skeleton, cartilage and viscera.6. Accumulation toxicity test: Until the 21st day, there were no dead animals. According to the standard of accumulation coefficient, K>5, so Sodium Benzoate is an antiseptic which has the function of lightly accumulation.So, the median lethal dose (LD50) peroral on mouse is 2100mg/kg.bw. Sodium Benzoate had potential mutagenic action, inheritance toxicity, and genetic toxicity. But there was no development...
