| The ubiquitin-proteasome pathway (UPP)is the principal mechanism for the degradation of short-lived proteins in eukaryotic cells.It is involved in a broad range of cellular processes,such as DNA repair,cell cycle progression,transcriptional regulation,antigen presentation and control of gene expression etc.Recently, studies including the mutation of the a-synuclein, parkin and UCHL-1 gene in familiar PD patients and cytoplasmic inclusions in sporadic PD patients have suggested that Ubiquitin-proteasome system is involved in pathology progress. Impairment of the Ubiquitin - proteasome system and the aberrant aggregated protein will result in cell neuronal dysfunction and death. That may be involed the mechanism of the absentance of dopaminergic neurons in PD.Objectiveâ‘ Establish the Parkinson's Disease model and identify it by the change of ultrastructure and DA value â‘¡Evaluate the learning and memory impairment in this rat model. â‘¢Examine the hHrdl's expression in the model orientationally and quantificationally and evaluate it's significance,which will provide a new theoretical foundation for clinic PD therapy.Methods â‘ Build up Parkinson's disease model rat by direct inject 6-OHDA into the nigra and the Medial forebrain bundle (MFB) with stereotactic technique.
