Objective:1 To explore initially the effect of fibronectin coating polyglycolic acid (PGA) on culture of chondrocytes in vitro, and find the best coating level;2 To compare the effect on the hydrophilia, absorptivity to chondrocytes and construction of tissue engineered cartilage of the PGA coated with different materials as cell scaffold.Method:This study consists of two parts. In the first part, free chondrocytes were isolated from rabbit articular cartilage by trypsinase and type II collagenase in order, amplified in vitro. The PGA scaffolds after sterilization were coated with fibronectin at different level of 50 μg/L ( group B ) , 100 μg/L ( group C), 200 μ g/L (group D ), 400 μg/L (group E ), 800 μg/L ( group F ) respectively, uncoated PGA scaffold as control group ( group A); the chondrocyte were seeded onto the six group scaffolds at the density of 4 × 10~7/ml, and then cultured in vitro for 4 weeks.
