Objective: To investigate the neuroprotective effects of topiramate on the changes of the nerve function deficiency scores ,SOD and MDA content in blood serum , the content of amino acids neurotransmitter (GABA, Glu ) in cerebral cortex and the content of GABAa recepector in ischemic territory of rats .Methods: Male SD rats were randomly assigned to sham operated group, ischemia-reperfusion group and TPM treated group. Then they were subjected to right middle cerebral artery occlusion for 2h and reperfusion for 24h. Rats in the treatment group were injected topiramate suspl ( 8mg/ml,80mg/kg) at the initiation of the artery occlusion and the reperfusion. The nerve function deficiency scores were measured. The infarct volume was evaluated through TTC staining. The concentration of MDA and SOD in blood serum were detected. Estimate the content of amino acids of cerebral cortex by the method of high performance liquid chromatography (HPLC) with fluorescent detection.
