Objective: We examined which were maximum tolerated drug dose (MTD) andoptimum dose by single intratumoral injection using 125I-UdR in nude mice xenografted with human bladder carcinoma. We examined the Eherapeutic Effect, body distribution radioactivity and safety of 125I-UdR after injecting intratumorally in nude mice xenografted with human bladder carcinoma; We evaluated the synergistic effect of combination 125I-UdR and Thymitaq in conjuncting 125I-UdR into DNA and the improving antitumous effect.Methods 1. Model of Bladder Carcinoma was found on BALB/cnu/nu mice, micewere randomly assigned to groups. In experimental groups (EG), after 125I-UdR was intratumorally injected with 61.67,123.33, 246.67 and 493.33 MBq / kg respectively, we viewed the therapeutic efficacy by measuring the change of volume and pathological change; To searching for MTD and optimum dose through analyzing SPECT scintigrams, radioactivity concentration in bodies of nude mice, peripheral blood cells (PBC), Hepatic function, renal function and the pathological change of normal tissues.The control group injected nothing. 2. Model of Bladder Carcinoma was found on BALB/cnu/nu mice, mice were randomly assigned to groups: (1) In experimental groups (EG), 125I - UdR was delivered by intratumoral injection at a dose of 123.33 MBq / kg in 40 mice; 7 d later, 15 mice were injected again; (2)20 mice were In Thymitaq group, Thymitaq(135mg/kg) was given by intraperitoneal injection, 24 h later 125I - UdR(123.33 MBq / kg ) was injected intratumorally; (3) The control group (25 mice) was intratumorally injected with Na125I (123.33 MBq/ kg); (4) All above groups were taken 10 mice for survival analysis in 100 d. Viewing the drug disposition by analyzing SPECT scintigrams and γ-counting, studying the therapeutic effect of all groups through pathology and survival.Results 1.
