Objective: To investigate anti-tumor effects of FAC, total effective components extracted from one medicinal herb.and its mechanisms in vivo and vitro.Methods: Inhibitory effects of FAC were investigated on the transplantable tumor models of H22 and L1210 in mice. Cytotoxicity of FAC on HL-60 and K562 cell line was confirmed by MTT and SRB assay in vitro. We used the immunohistochemical study, electron microscope observation, fluorescence microscope, flow cytometry, agarose gel, SDS-PAGE gel and western blotting analysis to further investigate the anti-tumor mechanism of FAC.Results: FAC had a great anticancer effect both in H22 hepatocarcinoma and leukemia in mice. FAC inhibited the proliferation of K562 and HL-60 cell lines, and markly decreased the amount of PCNA in H22 hepatocarcinoma cells. FAC could stimulate the immune function of mice implanted with H22 cells, and protected bone marrow of mice implanted with H22 cells from impaired effect of chemotherapeutants such as CTX. It could induce apoptosis in K562 cells. We also made it clear that FAC could down-regulate the amount of c-Myc protein and up-regulate the amount of p53 protein. It was also found that the amount of Fas protein in K562 cells was increased after treated with FAC, while the amount of FasL appeared no distinct change.Conclusion: Results of our experiment indicated that FAC had anti-tumor effects both in vivo and in vitro.
