Objective: To study the pathology change of animals with pulmonary fibrosis induced by bleomycin and effects of PNS on animals and associated mechanisms.Methods: Mice and rats were randomly divided into 6 groups : normal group,pulmonary fibrosis group,prednisolone group and three PNS treated groups(mice: 120 mg/kg,60 mg/kg,30 mg/kg;rats:60 mg/kg,30 mg/kg,15 mg/kg). Animal models of pulmonary fibrosis were reproduced by injecting bleomycin(5mg/kg) through trachea. Then each group gave corresponding drugs orally. On days 7,14,28 after drugs administration, the animals in each group were randomly killed and the lungs were harvested for histopathological studies,electron microscope studies and immunohisto-chemical determination of TGF-β,CollagenⅠandⅢ. Furthmore, assayed the level of SOD,MDA,HYP in lung tissue and blood serum.Results: (1) In the earlier period after bleomycin administration,alveolitis in model group is manifest(P<0.01). In the advanced stage,the pulmonary fibrosis is diffuse and serious. (2) On the 7th,14th,28th day, pulmonary index decreased after PNS administration(P<0.05,P<0.01). (3) PNS can significantly reduce inflammatory cells exudation and fibroblast proliferation in pulmonary interstitium and alveolar spaces on the 7th day after drugs administration(P<0.05,P<0.01). From the 14th day to the 28th day, the degree and range of fibrosis was attenuated(P<0.05,P<0.01).(4) PNS depressed collagen deposition and decreased the expression of TGF-βin lung tissue of pulmonary fibrosis model(P<0.05,P<0.01).(5) PNS can significantly increase the level of SOD and reduce the content of MDA in lung tissue homogenate and blood serum (P<0.05,P<0.01).(6) In the advanced stage, PNS decreased the level of HYP in lung tissue of pulmonary fibrosis model(P<0.05,P<0.01).Conclusions: 1,Mice and rats pulmonary fibrosis models were successfully reproduced...
