| bHLH(basic helix-loop-helix) transcription regulatory factors are important members of the transcription network during development. They are involved in many basic physiological processes such as neurogenesis, myogenesis, proliferation and differentiation of cells, the specification of cell subtype and the determination of sex.Mash1 gene, one of bHLH transcription regulatory factors is very important for neural development, neurogenesis and neural differentiation. Functional loss of Mashl gene resulted in abnormality of neural development, reduction of neuron and loss or reduction of certain neural subtypes. Nevertheless, the overexpression of Mashl gene prompted the neural differentiation of P19 cells, neural stem cells and embryonic stem cells. At present, plasmid vectors are commonly used in studying the overexpression of bHLH gene, but the transfection efficency is low. If Mashl gene is brought into stem cells in vitro or body or the targeted cells transfected Mashl gene are implanted into wound, it will persistently secrete recombinant protein in the local tissue and promote the neural differentiation and repair the damaged nervous system. For further studying the role of Mashl gene on neural development and neural differentiation, we constructed the Lentiviral expression vector of murine Mashl gene. |
