Mycotic keratitis is a severe disease which can badly damage vision of humankind. In recent years, incidence of mycotic keratitis keeps sustaining increases. Because of lack efficiency anti-fungi medication at present, early diagnosis is the key to design appropriate therapy. Conventional microbiological assay techniques for diagnosing microbial keratitis rely on culturing and specimen microscopy. But both have apparent deficiencies such as long time period and no Genus Identification of Fungal Pathogens. Another, different stains need different treat, so if we could detect the stains it would significant to treatment.The PCR assay was optimized using dilution of DNA template in sterile deionized water and the sensitivity of the multi-Polymerase chain reaction system be checked up. Clinical fungus strains were used to research the possibility of PCR systems clinical genus identification; To establish a rapid and simple extracting system of fungal DNA from clinical samples with fungal keratitis and evaluate the usefulness of the multi-PCR systems in comparison with the conventional mycologic methods in the diagnosis of fungal keratitis.Objective: To establish a method of fast diagnosis and species identified of mycotickeratitis by using multi-Polymerase chain reaction.Material and methods: Firstly, design four species primer.
