Objectives: To explore the expression patterns of NF-κB in two human medulloblastoma cell lines (UW228-2 and UW228-3) respectively, analyzing the relationship between NF-κB and growth, differentiation, apoptosis of medulloblastoma, investigating the impacts of resveratrol on the expressions of NF-κB and its biological implication. To further investigate the potential action mechanism of resveratrol on medulloblastoma cell lines and To provide a new laboratory evidence and theoretic basis for better treatment of human medulloblastoma.Methods: Human medulloblastoma cell lines UW228-2 and -3 were cultured in DMEM (Dulbecco's Modified Eagle's Medium) culture medium containing 10% Fetal Bovine Serum for 24h. Resveratrol (Sigma Inc.USA) was dissolved in demethylsulfoxide (DMSO) to a stock concentration of 5mM and diluted with culture medium to 100μM just before use. The cell proliferation situations of experimental groups were examined through Trypan Blue counting method. Sterilized cover slips were put in the dishes before cell seeding whenever cell shape observation or immunocytochemical analysis was performed. After above treatments, cover slips were collected and fixed with 100% cold acetone. Immunocytochemistry (ICC) and RT-PCR were used to analyze the expression levels of NF-κB.
