Objective: To study the pharmacokinetics of arsenic of compound nami-realgar in rats and compare the difference of pharmacokinetics between compound nami-realgar and compound realgar natural indigo tablets, for the purpose of development of a new drug.Methods: To collect serum samples after ig of drugs in rats at certain time, the concentration of arsenic in samples was determined by hydride generation-atomic fluorescence spectroscopy (HG-AFS) after microwave digestion. The pharmacokinetics parameters were calculated by 3P97 software. There were three doses groups in compound nami-realgar and one dose group in compound realgar natural indigo tablets.Results: (1) HG-AFS was proved to be sensitive, highly specific and precise, the recovery was 87.5% and the linearity is all good during 0.143ng/ml~10ng/ml. (2)After single ig doses of compound nami-realgar 35mg/kg,70mg/kg,140mg/kg to rats, the serum concentration-time curve was fit to one-compartment model. The main parameters: t1/2Ke(18.9881h, 21.5983h, 22.1156h respectively); AUC(5069.3480, 10650.3000, 23393.1100ng/ml·h respectively). Excepting AUC and Cmax, no significant differences were observed in the pharmacokinetics parameters among the three different doses. (3) After single ig dose of compound realgar natural indigo tablets 250mg/kg to rats , the serum concentration-time curve was fit to one-compartment model . t1/2Ke was 8.2116h, CL/F(s) was 0.0411L/kg/h, AUC was 5240.6480ng/ml·h. (4) There was a significant difference in pharmacokinetics parameters between high dose of compound nami-realgar and compound realgar natural indigo tablets.
