Objectives: To investigate the effect of recombinant adenovirus mediating human endostatin (rAD-ES) and traditional Chinese herbs on rats with collagen type II induced arthritis (CIA), and explore the mechanism of inhibiting angiogenesis and inflammation on rats CIA.Subject:Rat SD.Methods:The recombinant adenovirus carrying GFP-tagged human endostatin (rAD-GFP-ES) and rAD-GFP were amplified in HEK-293A cells, harvested and purified by centrifuge, and tittered by ultraviolet spectrophotometry. Infection efficiency was monitored with the expression of GFP protein.The model of rat CIA was induced by injection of intradermal collagen type II combined with complete Freund's adjuvant (CFA). On the second day after the injection, the therapeutic administration of rAD-GFP-ES (1×1011 pfu·kg-1·week-1×4 weeks) and traditional Chinese herbs(40g·kg-1·day-1×4 weeks)were performed to the rats, respectively. The rats in normal groups and model groups were administrated with rAD-GFP or PBS in the same dose. The mean arthritis index (AI) was scored every week.
