Objective: To investiage whether the eletransfection of human dendritic cells (DCs) with total RNA of hepatocarcinoma cells could induce the specific effective T cells, we oberve the changes of DCs'morphology and molecular expression between before and after eletransfection of human dendritic cells (DCs) with total RNA of hepatocarcinoma cells, as well as the changes of morphology and molecular expression between immature and mature DCs, then analysis the specificity that effective T cells kill target hepatocarcinoma cells.Methods: DCs obtained monocytes form human peripheral blood by density guadient centrifugation are incubated into immature DCs and mature DCs in the presence of cytokines in vitro. We use Wright-Gimsa staining, immunocytochemical staining, enzyme cytochemical staining and light microscope, invert microscope, electron microscope to observe the morphology and molecular expression of monocytes, immature DCs and mature DCs.To cultivate human hepatocarcinoma cells Bel-7402 in vitro.
