Object: To explore the effect of NF-KappaB in the onset of migraine by the observation of NF-KappaB expression in trigeminal nerve caudal core of the migraining rats.Methods: To divide the research to two parts. In the first part,72 Wister rats were random allocated into 6 experimental groups and 6 control groups(6 rats /group) .Glyceryl trinitrate(GTN) and isotonic Na chloride were subcutaneous injected into the experimental groups and control groups respectively. After 1, 2, 3, 4, 5 , 6 hours , the distribution of NF-KappaB positive staining cells in the trigeminal nerve caudal core of the migraining rats were determined with immunohistochemistry. In the second part,using Westernblot analytical method to semiquantitative analyse the expression of NF-KappaB proteinum in trigeminal nerve caudal core of the migraining rats in the corresponding times of the frist part.Results:Immunohistochemistry dyeing discover all of the control groups have the NF-KappaB nuclear positive reaction in caudal core of trigeminal nerve, and there is no obvious difference between each various time group, does not have statistics significance (P>0.05); the NF-KappaB nuclear positive reaction in all experimental groups increases more than control groups, otherwise within the experimental group, activation of NF-KappaB at 3rd and 4th hour is more than 1, 2, 5, 6 hours groups, have statistics significance (P<0.05). The Westernblot analysis confirms the same rule of NF-KappaB protein expression; 3 hours groups have statistics significance with other each group of comparisons (P<0.05), while 2, 3, 4 hours groups are more than 1, 5, 6 hours groups (P<0.05).Conclusions: The expression of NF-KappaB proteinum in trigeminal nerve caudal core of rats was reinforced after subcutaneous injecting GTN , it is showed that the reinforcement of NF-KappaB expression participate the onset of migraine.
