| Objective: To investigate the effects of Shenfu injection (SF) on the contents of NO,ET,6-keto-PGF1α and TXB2 in the canine model of myocardial ischemia reperfusion injury (MIRI) .Methods : 20 adult healthy cross bred canines were randomized into four groups: Control group, SF low dosage group(1ml/kg), SF middle dosage group(2ml/kg) and SF high dosage group(4ml/kg). Canines in all groups were injected SF(or NS.) before the operation under the same condition. The model of MIRI was established by ligating levo-anterior descending branch of coronary artery. The contents of ET, NO , TXB2 and 6-keto-PGF1α in plasma were measured at five time points: Pre-drug, ligation, 30 min after ligation, 1 hour and 2 hour after reperfusion respectively and the contents of NO,ET,6-keto-PGF1α and TXB2 were analyzed. Myocardial tissue was observed by light and electron microscope after operation.Results : Compared with the control group , SF high dosage group reduced the levels of NO,ET and TXB2 , promoted the expression level of 6-keto-PGF1α, and regulated the equilibrium between 6-keto-PGF1α and TXB2 (P<0.05). Light and electron microscope showed evidently that the pathological changes of myocardial tissue were more slight in SF high dosage group than in Control groupConclusion: SF injection has good protective effects on MIRI, the mechanism of which relates to its function of decreasing the levels of NO,ET and TXA2 , promoting the expression level of PGI2, and regulating the equilibrium between PGI2 and TXA2. |
