Effect Of Berberine On Intimal Hyperplasia And Vascular Remodeling After Balloon Withdrawal Injury Of Rabbit Carotid Artery

ObjectiveTo observe the effect of Berberine on the levels of Endothelin-1 ( ET-1), Nitric Oxide (NO), Platelet Derived Growth Factor ( PDGF ), Transfer Growth Factor-pi(TGF-β1 ) and their effects on Intima Hyperplasia and Vascular Remodeling after Balloon Withdrawal Injury of Rabbit Carotid Artery, and evaluate the mechnism of preventing restenosis. MethodsFourty Japanese rabbits were allocated randomly into 4 groups: Artificial group( n=7 ), model group(n=11), Berberine group( n=11)and Simvastatin group( n=11). We made balloon endothelium denud-ation in Carotid Artery of rabbits in the later three groups in the condition of anesthesia, and they were injected respectively with 0.9% Sodium chloride, Berberine and Simvastatin liquid (2.5mg/kg·d) . After 15 days, the levels of ET-1 ,NO and PDGF, TGF-β1 of peripheral blood were detected by Nitrate Reductase, Radioimmunology, Enzyme Linked ImmunoSorbent Assay( ELISA) . At the same time, we observed the morphologic change of injured artery and took photos, then Intimal Thickness(IT), Intimal Area(IA),Luminal Area (LA), Internal Elastic Lamina (IEL), Medial Area (MA) and External Elastic Lamina (EEL) of all groups were measured with the system of computer photo analysis, IA/MA and Intima Hyperplasia Index (IHI) were computed. Results(1) Generic Condition: All groups appeared bluntness, twitch, coat-turbulence, lackluster, shrink-rickets, few-movements , scour, few-dining or emotional et al, and found dead rabbits from 5th day. The symptoms of berberine group, simvastatin group and artificial group were lighter than model group, and berberine seems have a direction of decreasing the rate of death.( 2 ) Blood Index: After 15 days, the elevation of NO of berberine group were unconspicuous compared with artificial group but significant compared with model group and simvastatin group(P<0.01 ); the ET-1 levels of berberine group , simvastatin group and artificial group were markedly lower than model group (P<0.01), but 3 groups have not obvious difference with artificial group( P > 0.05 ); the levels of ET-1, PDGF and TGF-β1 of Berberine group,Simvastatin group and artificial group were significantly decreased ( P<0.01 ) compared with model group, but 3 groups have not obvious difference ( P > 0.05 ) .( 3 ) Pathology: The intima of artificial group were glossy without incrassation, we can see flat endothelium cells under intima, Internal Elastic Lamina were full, nucleolus of smooth muscle cells were shuttle; The intima hyperplasia of model group were grievous mostly, the repairing of intima of was not full, Internal Elastic Lamina was discontinuous, the surface was not glossy, and we can see a lot of smooth muscle cells moving and hyperplasia from media to intima, the cells were turbulence, with a...
