Objective: To establish animal experimental model of human Non-small-cell-lung-cancer (NSCLC) cell dormancy in nude mice, and to verify the effect of nm23-H1 gene on promoting dormancy of tumor cell. The model will be used for investigating the mechanism and therapeutic method of tumor dormancy.Methods: L9981-nm23-EGFP is a human NSCLC cell line transfected with plasmids pEGFP-nm23-H1, an eukaryotic expression vector which harbor the gene of nm23-H1, but L9981-EGFP is a human high-metastatic NSCLC cell line constructed by transfecting the control eukaryotic expression vector of pEGFP into L9981 cell line, which does not express the gene nm23-H1. L9981-nm23-EGFP lung cancer cells or L9981-EGFP lung carcinoma cells were inoculated into the nude mices by tail vein injection to establish the experimental lung metastasis model of NSCLC. The former was considered as experimental group (L9981-nm23-EGFP), and the latter as control group (L9981-EGFP). All mice were killed by neck-broken method after 8 weeks.
