Objective: To investigate the influence of 3,4-dibromo-5-hydroxy-2(5H)- furanone, 4-bromo-5-(4-methoxyphenyl)-3-(methylamino)-2(5H)-furanone and3,4-dibromo-5,5-bis(4-methylphenyl)-2(5H)-furanone to the biofilm formation of Staphylococcus aureus(SA), Staphylococcus epidermidis(SE) and Escherichia coli(EC) on the surface of cardiovascular biomaterial.Methods: 288 slices of PVC which often be use to make cardiovascular biomaterial were average divided into four groups. Three groups of the PVC slices were coated on 3,4-dibromo-5-hydroxy-2(5H)-furanone; 4-bromo-5-(4-methoxyphenyl)-3-(methylamino)-2(5H)-furanone and 3,4-dibromo-5,5-bis(4-methylphenyl)-2(5H)-furanone respectively as the experimental groups, the other group was control group. Each of the four groups of PVC slices was average divided into three subgroups again. The subgroups of PVC slices were separately putted into the culture flasks respectively contained culture fluid of SA, SE, EC with the bacterium concentration of 10~5CFU/ml. After cultivated 6,12,18, 24 hours, we used Confocal Laser Scanning Microscope (CLSM) to measure the depth and the density of the biofilms on the extracorporeal circulation set tube. On the other hand, we used environmental scanning electron microscope (ESEM) to observe the configuration of the biofilms on the surface of the extracorporeal circulation set tube. Results:1. Staphylococcus aureus group: The depth and the density of the biofilms were significantly increased on the surface of the PVC material coated with 3,4-dibromo-5-hydroxy-2(5H)-furanone compared with the control group(P<0.05). |
