Immunological Efficacy Of HSP70 Peptides To Glioma

Objective: Purify heat shock protein70-associated tumor peptides( HSP70-PC) from G422 glioblastomaglioma, immunize mouse with HSP70-PC via s.c, investigate its immuno-protective effect against glioma on mouse in vivo. Methods: HSP70-PCwere acquired from G422 glioblastomaglioma. After immunize-therapy of mouse with HSP70-PC(10μg) and cell lysates (10 μg) vaccine respectively, they were inoculated with G422 tumor cells after two weeks . The progression of tumor and life span ofmice were test. Results: In an mice model of intracranial G422, the rate of tumorformation in normal saline (NS) group, the cell lysates group and HSP70-PC group was 100% , 100%, 83.3% respectively, the fist time of tumor formation was 10 d, 12 d, 15 d respectively, the lifetime was (16. 00±0.58) d, (18.70±1.33) d,(25.33 ±1.86)d respectively.In an mice model of G422 inoculated via s.c, the rate of tumor formation in NS group, the cell lysates group and HSP70-PC group was 100%, 100%, 33.3% respectively, the weight of tumor was (6.00±1.67) g, (2.30 ±1.23) g, (0.12±0.20) g respectively, the volume of tumor was (4.33±0.44) cm~3, (2. 65±0.31) cm~3, (0.17±0.32) cm~3 respectively, and flow cytometric analysis of the blood of mice showed that lymphocytes was (32.65±9.06) %, (51. 30 ± 7. 72)%, (67. 42±6. 00)% respectively, CD3~+CD4~+T lymphocytes was(14. 28 ± 6. 75) %, (17.17±5. 04) %, (22.5±5.18) % respectively, CD3~+CD8~+ T lymphocytes was (4.30 ± 1.97) %, (7.06 ± 2.20) %, (11.75 ± 3.57) %respectively., Conclusion : HSP70-PC can induce response of CTL and prolong the lifetime of mice, which have obviously immuno-protective effect against glioma.
