| The purpose of this study was investigated the characteristics of apoptosis of osteocyte induced by Porphyromonas gingivalis and the inflammatory pathological change during the bacterial-induced tissue destruction . One hundred twenty male Kunming mice which were divided into experimental and control groups. The mice in experimental group were inoculated with Porphyromonas gingivalis in the scalp of mice . The control mice with inoculated with 1×PBS. The animal models were killed at 0,1,3,5,8and 12days after inoculation .The inflammatory pathological change was observed with hematoxylin and eosin (HE). Tartrate resistant acid phosphate (TRAP)assay on tissue was perform as describe in osteoclasts . Apoptosis of osteocyte were examined by immunohistochemical staining and Tdt-mediated dUTP nick end labeling(TUNEL) during series time of after inoculation. The results showed that after the inoculation of P.gingivalis, the inflammation and destruction in connective tissue were observed. The number of apoptosis osteocyte were detected increased greatly with inflammation which was significiantly difference between control group and experimental group . These results suggest that osteocyte apoptosis is induced by theperiodontal pathogen------Porphyromonas gingivalis and support the hypothesis thatosteocyte apoptosis mechanisms could be implicated in the inflammatory process associated with osseous tissue destruction and the bone loss which higher than new reparative bone formation . |
