Achyranthes Bidentata is one of ox knee category plants,widely distributed in many places of China except for Northeast China, and known as a kind of Chinese traditional tonic herb.The constituents in Radix Achyranthes Bidentata include triterpenoid saponins, polysaccharides,steroids,etc.Recently the main research has been centered on polysaccharides which was systemly introduced on chemical structures and pharmacology.the reports on triterpenoid saponins have been less than polysaccharides.Now triterpenoid saponins has many important biological activities,such as anti-fertility, relieve-pain, antibacterial, immune adjustment and anti-inflammatory activities,but its officinal research is not in-depth and not systemic.The experiment aims at selecting the anti-fertility component or sect from Achyranthes Bidentata and lays a foundation for exploiting Achyranthes Bidentata to a new and effective Botanic sterilant against pest rodents .1.The extraction and isolation of total saponins of achyranthes(TSA) from Achyranthis bidentatasaponins from Achyranthes bidentata are triterpenoid saponins wholly. We applied traditional phytochemistry separation methed and TSA were extracted by cool ethanol methods from powder of Achyranthes bidentata .The extraction rate of gum from the cool leach method was 4.26%. The gum was extracted by aether ,acetal and n-butanol saturated with water in succession ,then pured by aether-deposit and column chromatography. The extraction rate of TSA was 1.7%.2. The determinant and quantitative test of TSA from Achyranthes BidentataTSA from Achyranthes Bidentata contained mostly acidic saponins without steroidal saponins. It concluded by the results of the color reaction ,suds reaction and the thin layer chromatography. The structures mainly contained triterpenoid saponins which sapogenin was oleanolic acid.A TLC-scan method was developed for determination of oleanolic acid in roots of Achyranthes bidentata .The key experimental conditions were as follows:saponin hydrolysis at 100℃with 1mol/L sulfuric acid for 7 h,TLC separation on a home-made silica gel G plate with chloroform-ethyl-ether-methanol(30:10:1)as a developer,visualization at 100℃for...
