| Purpose: Ginsenoside Re had been proved one of effective components of ginseng in medical and pharmacological studies. It was believed that Ginsenoside Re content formed 23% of total Ginsenoside, it was the most part in total Ginsenoside. Therefore, it should be the representative of Ginseng. However, fewer pharmacological studies on Ginsenoside Re had been carried out and the effects of it on Shock had not been reported. So this research would evaluate the effect of Ginsenoside Re on Shock through different animal models, in order to provide preclinical pharmacological experimental data.Methods: In superior mesenteric artery occlusion(SMAO) of rats model was established by clamping superior mesenteric artery (SMA) for 2 hours and then reperfusing for another 2 hours. During the whole process mean artery pressure (MAP) and heart rate(HR) were recorded. The blood samples were collected before and 2 hours after clamping respectively, to determine serum GOT, GPT, LDH, GLU, CK, BUN , Cr and NO. Intestine, lung, heart, kidney and liver tissue samples were also collected after reperfusion 2 hours, and (1) protein concentration of bronchia alveolus lung fluid [BALF], (2)HSP70 expression in heart and kidney (3) histology pathology and (4) oxidation injury (MDA and GSH-PX) in above tissues were determined.Hemorrhagic shock(HS) of cats was initiated by blood withdrawal till the MAP reached 40 mmHg within 15 min and maintained this state for 90 min, then model animals were resuscitated by transfusion of tested substances and the shed blood. Then, every group cats were killed 2h after transfusion, blood samples were collected to deternmin MDA and LD. In addition, blood gas analysis ,WBC and Hb were detected. Then histologis assay was performed.The evaluated marks of rats' scald shock model were Hct, LDH and CK-MB. And mice insulin shock model was evaluated through recording the alive time and convulsion latency of animal after Ginsenoside Re injection.Result: Ginsenoside Re decreased the morality of SMAO rets and the GOT, GPT, LDH, CK, BUN ,NO and Cr, the GSH-PX activity was increased and MDA content was decreased. Moreover, the expression of HSP 70 in heart and kidney were up-regulated and the protein content in BALF was inhibited after Ginsenoside Re treatment. |
