| [Objectives]Reactive hyperplastic colloidal scar after spinal cord injury(SCI) is the key to block the regeneration of SCI. Chondroitin sulfate proteoglycans(CSPGs) is the main element of colloidal scar and forms a chemical barrier in front of axonal regeneration.The purpose of this experiment is to study:â‘ making and nursing of the model of rat spinal cord transection; â‘¡the effect of chondroitinase ABC on local scar after SCI;â‘¢the effect of chondroitinase ABC on the restore of function of spinal conduction. [Methods]â‘ made the model of rat spinal cord transection, divided them into SCI group(A), SCI nursing group(B), SCI treatment group(C) and analyzed the effect of nursing on rat mortality and survival span; â‘¡used the grading method of BBB to value the behavial changes 1, 3, 7, 14, 21 days after rat injury; â‘¢observed the dissolving effect of chondroitinase ABC on CSPGs through cryostat sections and immunofluorescent histochemistry 4 weeks after rat injury; â‘£observed neural regeneration through HRP retroantergrade shows 8 weeks after rat injury. [Results]â‘ there is significant difference of living quality and mortality among three groups, group C is better than B and group B is better than A(P < 0.05); â‘¡difference of BBB grades among three groups is signifcant, group C is better than B and group B is better than A(P < 0.05); â‘¢horizontal cryostat sections from the lesion site were immunolabeled using two antibodies against CSPG., chondroitnase ABC reduces the anti-CSPG-carbohydrate epitope immunoreactivity, but not the CSPG-protein core pitope; â‘£At the positive of spinal cord,the HRP retroantergrade shows that there are a few labelled cell bodies and axons of neurons in group C 8 weeks after SCI. |
