| Objective: To detect the influence of Ad-hBMP-2 toward osteoblasts in vitro. To provide experimental evidence and theory guidance when Ad-hBMP-2, osteoblasts and compound biomaterials are used to treat clinical bone defect or reshape bones.Methods:1. HEK (human embryonic kidney line) 293 cells, as packaging cells, were used for culture of recorabinant adenovirus vectors containing hBMP-2 gene. Adenovirus vectors containing hBMP-2 gene were reproduced and purified by cesium chloride density gradient ultracentrifugation. Biological activity identification was carried out using infectious dose 50.2. Cultured osteoblasts form cranial bones of newly born New Zealand rabbits are used for cell model. The difference between transfected osteoblasts and raw osteoblasts in shape, growth, reproducement, function was investigated through phase-contrast microscope observation, tetrazolium salts chromatometry detection, alkali phosphatase activity measurement , immunocytochemistry detection, RT-PCR detection and dyeing with alizarin Bordeaux.Results:... |
