[Objective]To investigate the effect of ethanol withdrawal on excitatory amino acid neurotransmitter in hippocampus DG of conscious rats, and study the effects of Topiramate on ethanol withdrawal .[Method] Fifty three-month-old rats were randomized into Control group(n=10) and Experiment group(n=40). Experiment group were orally exposed to 50% ethanol(10ml·kg-1-d-1,ig) for 1 week(n=10), 2 weeks(n=10) and 4 weeks(n=20) and TPM(80mg·kg·d-1, 1,ig)was given to 4 -week ethanol group starting in ethanol withdrawal. Control group were exposed isovolume saline. The microdialysis technique and high-performance liquid chromatography was used to quantify glutamate and asptamate in hippocampus DG with each rat at 6h, 30h, 48h and 72h during the ethanol withdrawal. At the same time , the ethanol withdrawal sydromes were observed.[Result](1)The glutamate and asptamate of 2 -week ethanol group and 4 -week ethanol group at 30h and 48h were significantly increased than that of 6h during ethanol withdrawal (p<0.05) .(2)There is no significant difference between Control group, 1-week ethanol group.(3)The glutamate and asptamate Topiramate group were not increased at 30h and 48h than that that of 6h during ethanol withdrawal (p>0.05).
