| The conventional method to evaluate disinfection effect is to observe bacterial existence and count of viable bacteria. This qualitative and quantitative analysis is very exact, but at the same time it' s time-wasting and exertive. So seeking rapid method to assess bacterial existence and number is attractive. This study carried out bioluminescence-based assays for rapid evaluation of disinfection effect.Luciferase is used broadly in gene engineering. Especially as reporter gene, luciferase is important in analyzing gene activity, gene localization and spacial expression. Luciferase catalyze the followingbiological chemical reaction:Luciferin + ATP + O2→|(Luciferase, Mg2+) Oxyluciferin+AMP+pp1+CO2+LightIn this enzymatic reaction, if reactant is excessive, then the quantity of the light produced from the reaction is proportional to the quantity of the luciferase.In this study, we constructed B. coli recombinant expressing luciferase at first. Then this E. coli recombinant was used in suspension quantitative germicidal test to several disinfectants and ultraviolet.
