Study Of Effect On Percentage And Function About CD4～+CD25～+T Cells In Unexplained Recurrent Spontaneous Abortion

ObjectiveTo investigate the effect on percentage and function about CD4~+CD25~+ T cells (CD4~+CD25~+Treg) in unexplained recurrent spontaneous abortion(URSA).Methods1.Proportion measurement: The proportion of CD4~+CD25~+Treg in peripheral blood was measured by double-label flow cytometric analysis . The samples were taken from 15 URSA women, 15 normal non-pregnancy women and 13 normal pregnancy women.2.Function measurement: CD4~+CD25~+Tr cells and CD4~+CD25~-T cells were extracted from peripheral blood lymphocytes by the microbeads separation . Flow cytometry measured the purity of CD4~+CD25~+Tr cells and CD4~+CD25~-T cells. The suppression function of CD4~+CD25~+Tr cells on CD4~+CD25~-T cells was accessed by the growth inhibition in vitro.ResultsThe proportion of CD4~+CD25~+Tr cells was decreased significantly in URSA women (6.88±1.81)%than that in normal non-pregnancy women[(10.8±1.05)%] (P< 0.05) and normal pregnancy women[(11.21 + 1.40)%] (P<0.001) .In function, the suppression rate of CD4~+CD25~+Tr cells was decreased significantly in URSA women(75.0±5.89)% than that in normal non-pregnancy women[(89.34±3.57)%] (P...
