[Objective] Detect the expression of VEGF-C and VEGFR-3 in the human breast carcinoma and benign disease by tissue chips and research its relationship to regional lymph node metastasis.[Method] 1.109 cases primary breast carcinoma and 30 cases mammary benign disease specimens were collected and paraffin imbedded.2. The tissue chips were consisted of cores ( spot diameter =0.6mm) that taken from the centre and borderline of carcinoma and distant normal mammary gland of each breast cancer, and mammary benign disease specimens. 3. The specimens of tissue chips were detected for the expression of VEGF-C and VEGFR-3 by Immunohistochemical method. All the results were statistically analyzed with clinicopathologic factors.[Result] 1. The tissue chips containing breast carcinoma and benign disease specimens were successfully constructed, and got well effect in Immunohistochemical detection. 2. The positive rates of VEGF-C and VEGFR-3, and the number of VEGFR-3 positive vessels in the centre and borderline of carcinoma and distant normal mammary gland were 69.4%, 39.8%,12.42±6.4, 69.1%, 43.5%,9.60±6.4, 52.9%, 13.2%, 2.29±1.6, but not in benign disease specimens. 3. With the increase of VEGF-C and VEGFR-3 protein level in the centre and borderline of carcinoma, the number of VEGFR-3 positive vessels significantly increased (p<0.05). 4. Most VEGFR-3 positive peritumoral vessels had wide lumena, whereas most VEGFR-3 positive intratumoral vessels had fissure or collapsed lumena. 5. The positive index of VEGF-C and VEGFR-3, and the number of VEGFR-3 positive vessels in lymph node metastases group were significantly higher than that of no metastases (p<0.05). 6. There were no significantly correlation between the over expression of VEGF-C and VEGFR-3 and clinicopathologic factors such as ER, PR, P53, c-erbB-2, and the size, number, clinical stage and pathologic type of breast carcinoma.[Conclusion]...
