Isolation And Structure Determination Of Antibiotics From A Micromonospora Strain: C5308

Three blue compounds were isolated from the mycelia of Micromonospor nigra C5308 by using the methods of parall-bacteria and spermatogonium as the screening model. The mycelia were extracted with acetone. The fat-soluble part of the concentration was chromatographed on columns of silica gel. C5308-a, C5308-b and C5308-C were obtained from the mixture of the blue eluate, by means of preparative TLC and ODS chromatography.The structure of two of them, named C5308-C and C5308-b, was established through spectral analyses as Dynemicin M and Dynemicin P, which contains a anthraquinone portion and chemical named as 9,8,14-[1]Butanyl[4]ylideneanthra [1,2-b]benz[f]azocine-5,16,19-trione, 8,9,14,15-tetrahydro-1,4,6,8,17-pentahydroxy -20-methoxy-18-methyl-, [8R-(8a,9b,4b,17S*,18S*)]- and9.8.14-[1]Butanyl[4]ylideneanthra[1,2-b]benz[f]azocine-5,16,19-trione, 8,9,14,15-tetrahydro-1,4,6,8,17,20-hexahydroxy-18-methyl-, [8R-(8a,9b,14b,17S*,18R*)]-. Complete assignments of ¹H and ¹³C spectra of Dynemicin M and Dynemicin P (C5308-C and C5308-b) were achieved by 2DNMR spectra, such as HMBC, HMQC.These three compounds have certain relationship with Dynemicin A, the great antitumor antibiotic that contains an enediyne portion.
