Object: In order to investigate the mechanism of Ligustrazine Hydrochloride as a taboo for pregnant women.Methods: The pregnant mice were treated intragastrically with Ligustrazine Hydrochloride (10mg/d, 2mg/d or0.4mg/d) for six days from the first day of pregnancy and the control group was treated with saline only. At the seventh day of pregnancy, uterus and spleen were removed and blood was collected. The progesterone level in serum was measured by RIA. The splenocyte were cultured in vitro, and the amount of splenocyte was detected by MTT. The expression of the proteins of TGF-β1 and TGF-β1RI were detected by Immunohistochemical staining and the expression of TGF-β1 mRNA was detected by in situ hybridization histochemistry.Result: The level of progesterone in serum distinctly increased with different concentration of Ligustrazine Hydrochloride as compared to the control group. As to the cultured spleen cells after treated with 10mg/d and 2mg/d of Ligustrazine Hydrochloride, the cell number sharply increased. Ligustrazine Hydrochloride can weaken the expression of TGF-β1, but had no effect on the expression of TGF-β1RI in none of the three experimental groups. The expression pattern of TGF-β1mRNA was similar to the TGF-β1. TGF-β1mRNA was distinctly reduced at the dose of 10mg/d of Ligustrazine Hydrochloride.Conclusion: Ligustrazine Hydrochloride can enhance the multiplication of spleen cells may be by reducing the expression of TGF-β1. Progesterone as a immune inhibitor exert its function-depending on many cytokine include TGF-β1.
