In this thesis, a novel chemiluminescence(CL) system of K2MnO4-luminol for determination of metal ions was presented. And based on this, an in-vivo and real-time analytical system for monitoring the metabolism of free lead (Ⅱ) and nickel(Ⅱ) in rabbit blood was developed. That was, the analytical system was composed of microdialysis sampling, on-line separation and chemiluminescence detection. Furthermore, lead (Ⅱ) and nickel( Ⅱ) in the rabbit liver, kidney, heart and urine were also determined by flame absorption spectrometry.This thesis is made up of the review and research section.The review includes mainly two parts:Firstly, abortion and metabolism of lead and nickel in the body and determination methods of them are introduced respectively in this part;Secondly, the basic principles of microdialysis sampling and its quantitative determination and application are summarized here.The research section contains three subunits.Subunit 1, the study of a novel CL system: M-K2MnO4-luminl (M presents metal ions)A novel CL system of M-K2MnO4-luminol was developed. Potassium manganate solution was prepared firstly. Then it was found that the reaction between K2MnO4 and luminol could produce chemiluminescence and the CL intensity could be enhanced or inhabited by some metal ions. For example, Cu(Ⅱ) would inhabit the chemiluminescence reaction of K2MnO4 and luminol greatly, whereas Pb(Ⅱ), Ni(Ⅱ), Mn(Ⅱ), Ag( Ⅰ ), Mg(Ⅱ) and so on could enhance the chemiluminescence intensity. And based on these observations, the CL system allowed to determine 5×10-8 9×10-7 g/mL Pb2+, 1×10-10 1 ×10-8 g/mL Ni2+, 3 ×10-7 1 ×10-6 g/mL Ag+, 1×10-9 1 ×10-8g/mL Mn2+, 2×10-7 8×10-7 g/mL Mg2+ with the detection limits of 2×10-8 g/mL, 7×10-11 g/mL, 8×10-8 g/mL, 2×10-10 g/mL, 2×10-8g/mL, respectively.Submit 2, the study of in vivo and real time monitoring the metabolism of unboun lead (Ⅱ) in rabbit blood with microdialysis sampling combining FIA-CL detection...
