Objective: In older to study antitumor mechanisms of tumor-draining lymph node (TDLN) cells in vitro and in vivo, we carried out several experiments. It is helpful to improve the cultural method of TDLN cells and provid new ideas for tumor biotherapy.Methods:1 We prepared the single cell suspension of TDLN cells from lung cancer patients.2 We prepared the single cell suspension of cancer tumor.3 We induced TDLN cells with different stimuluses: We put the single cell suspension of TDLN cells (1×106/ml) into 24 shadow mask, and divided them into three groups, every group induced by a different stimulus. The first group is induced by IL-2 (175U/ml); The second group is induced by IL-2 (175U/ml)+ tAg (50μg/ml); The third group is induced by IL-2 (175U/ml)+GM-CSF (500U/ml)+IL-4 (100U/ml)+tAg (50μg/ml).
