Objective: To find a better method to deal with interstitial lung disease, we investigate interferon-gamma combined with methylprednisolone how to influence human embryonic lung fibroblast on proliferation, collagen synthesis and the expression of transforming growth factor-beta one protein and mRNA.Methods: Human embryonic lung fibroblasts were cultured in the environment of 37℃, 5% CO2, saturated humidity and RPMI-1640 which supplemented with 10% new born bovine serum medium, 100 units/ml penicillin and 100ug/ml streptomycin. We performed 2×4 factorial experimental design. There were several groups, which are control group, alone using methylprednisolone group, alone using different concentration interferon-gamma groups and different concentration interferon-gamma combined with methylprednisolone groups. Exponentially growing cells were preincubated for 48 hours before they were harvested and used for the analysis of the following items:The microculture tetrazolium (MTT) assay was used to measure the inhibition ratios of human embryonic lung fibroblasts: The cells were digested and transferred into 96-well microtiter plates (1×105/ml/well). Every sample's...
