AIM: Arrhythmias remain a leading cause of mortality and morbidity, claiming a large number of victims every year. exporing the mechanism of the arrhythmias and seeking the safe and effective antiarrythmic therapy become the research hot spots. Ibutilide, as a newer class III antiarrythmic agent, is effective in the atrial and ventricular Arrhythmias. At the meantime, ibutilide also brings some side effects, mainly Torsade de pointes(Tdp), which have closely relationship with L-type Ca2+ current. In physiological situation, L-type Ca2+ current participates the 2 period of repolarization in fast responsive myocardial cells, affects the action potential duration (APD) and the effective refractory period (ERP). The aim of the present study is to character the effects of ibutilide on L-type Ca2+ current of rabbit cardiomyocytes and to explore the electrophysiologic bases of antiarrythmic and proarrhythmic potential of ibutilide using patch clamp techniques.Methods: Front wall midmyocardial of left ventricular were isolated from New Zealand white rabbits ( either sex, body weight 2.0 to 2.5 kg).
