Objective: The immunohistochemical expression of APC and Ras proteins, and the DNA content in colorectal adenoma were investigated..The relationship between them with the mechanism that colorectal adenoma make progress were explored .To provide a biological markers for early diagnosis of colorectal adenoma .Methods: 116 colorectal adenomas,34 colorectal cancers and 11 colorectal normal mocosa specimens at SHANXI PROVINCIAL PEOPLE'S HOSPITAL from 2004 to 2005 were collected to carry on histopathological classification .The immunohistochemical expression of APC and Ras proteins were investigated by Elivision two-step method .The DNA content was detected by the flow cytometry . The relativity of them as well as the relation between them and adenoma or adenocarcinoma malignant degree were analyzed.Results :①The degree of atypia hyperplasia and canceration among tubular adenoma ,tubulo-villous adenoma and villous adenoma were statistically different(p<0.05). The canceration rate of diameter bigger or equal to 2cm was higher than diameter smaller to 2cm (p<0.05).②There were 81.8% of normal mocosas ,41.4% of adenomas and 23.5% of adenocarcinomas positive for APC expression .The expression of APC was consistently higher in normal mocosa than in adenoma and adenocarcinoma (p<0.05).The expression of APC between adenoma and adenocarcinoma weren't statistically different.The expression of APC were respectively 61.1%,40.8%,20.0%,18.8% in the four grade groups.APC expression was significantly higher in low-grade dysplasia adenoma than high-grade dyplasia adenoma or canceration adenoma(p<0.05).③There were 9.1% of normal mocosa,49.1% of adenomas and 73.5% of adenocarcinomas positive for Ras . Ras was statistically higher in adenocarcinoma than in adenoma while in adenoma than in normal mocosa(p<0.05). The expression of Ras were 22.2%,53.1%,73.3%,75.0% in the four grade groups.Ras expression was significantly lower in low-grade dysplasia adenoma than the other three groups adenoma .④The polyploidy rate of normal mocosa , adenoma or adenocarcinoma was respectively 0%,42.1%, 77.3%.The polyploidy rate of the three groups were statistically different(p<0.05).The polyploidy rate of four groups were individually 0, 25.0%,70.0%, 75.0%.
