| Objective:To study the effect of intestinal endotoxemia dynamic change on NOS express in rats lungs in the forming process of Hepatopulmonary syndrome (HPS).Methods: Twenty four male Wistar rats were randomly divided into four groups:control group ,fourth week group,sixth week group and eighth week group.The rats in control group were fed with regular food and those in cirrhosis group with food consisting of maize flour ,cholesterol(5%w/w) and alcohol(30%).The cirrhotic rats were administered with carbon tetrachloride (5ml/kg)on the first day,then administered with 40% carbon tetrachloride (diluted by maize oil,3 ml/kg) subcutaneously every four days. Rats were studied at the end of the fourth week,sixth week and eighth week respectively. There were 6 rats at least as study subjects in each group. Endotoxin,ALT,TNF-α.,ET-1 in plasma were detected by biochemistry and radioimmunoassay. Liver and lung tissue samples were collected for detected by pathology,immunohistochemical staining and RT-PCR.Result:1: The Dynamics research of IETM at various time points with the HPS of rats.The levels of plasma endotoxin were increased at the end of the 4th week in the forming process of HPS. The levels of plasma endotoxin were reached the peak at the end of the 6th week,and slightly dropped at the end of the 8thweek.2: The dynamic changes of NOS protein express and gene express in rats lungs in the forming process of HPS.2.1 The dynamics research of NOS protein express results indicated that the levels of iNOS protein expression in pulmonary architecture were increased gradually in the forming process of HPS, which achieves the peak at the end of the 8th week; and there were similar result with the eNOS protein express.2.2 The dynamics research of NOS gene express results indicated that the levels of iNOS gene expression in pulmonary architecture were increased gradually in the forming process in HPS; and there were similar result with the eNOS gene express, which were all consistent with the protein expression results.3: The effect of IETM on NOS express in rats lungs in the forming process of HPS,The results indicated that the levels of plasma TNF-αwere increased at the end of the 4thweek, and decreased at the 6thweek compare with 4thweek, reached the peak at the end of the 8th week;ET-1 were decreased at the 4thweek, increased at the 6thweek compare with 4thweek, reached the peak... |
