Objective:The objective of this paper is to quest for the normative method of building Kidney-yin deficiency rat model with thyroid, and to provide a first step standard of model evaluation as well, which can offer convenience for the study of Chinese medicine, and supply references to standardization work of animal model of traditional Chinese medicine. Method:1. theoretical research: To choose evaluation index of Kidney-yin deficiency animal model.1.1 To tally up clinical diagnostic standard and objective index of Kidney-yin deficiency syndrome by consulting correlative literatures, including the symptom and the objective evaluation index.1. 2 To draw-up the first step evaluation index of Kidney-yin deficiency animal model on the base of the above research results.2. experimental research:2.1 experiment â…  : To select preferable method of making Kidney-yin deficiency model in male SD rat with ThyroxineIn this experiment, chose SPF male SD rat, 200-240g, used dosages according to the following:6.25mg/kg, 12. 5mg/kg, 25mg/kg, collected blood on 14th, 21th, 28th respectively, then test the numerical value of Cort, T, E2, T3, T4, cAMP, cGMP in the blood, and compared with the blank group in order to select preferable dosage and time consequently.2.2 experiment â…¡ : To select preferable method of making Kidney-yin deficiency model in male SD rat with Thyroxine and ReserpineIn this experiment, chose SPF male SD rat, 200-240g, used dosages according to the following:6.25mg/kg, 12. 5mg/kg, 25mg/kg, collectedblood on 6th, 10th, 14th respectively, then test the numerical value of Cort, T, E2, T3, T4, cAMP, cGMP in the blood, and compared with the blank group in order to select preferable dosage and time consequently.2.3 experiment HI: To observe the cardinal symptom affected by Thyroid In this experiment, made Kidney-yin deficiency rat model which hadestablished in the experiment I and experiment II, observing the general response affected by Thyroid at the same time. All the rats were tested cold endurance capacity, provoked degree by snatching, run distance, autonomic activity, genito-function(male), etc. Results would be compared with the blank group. Through the symptoms observed supplement the objective index of experiment I and experiment II, and modify the symptoms system of Kidney-yin deficiency rat model farther.2.4 experiment IV : To observe the curative effect treated by Liuweidihuang Wan.The rats were divided into three groups randomly: model group, Liuweidihuang Wan group and blank group. Model group and Liuweidihuang Wan group were took solution as before, and organic pathology, while each Liuweidihuang Wan group' s rats were treated by Liuweidihuang Wan solution 4.5ml everyday in the same time , the blank group took saline. During the course of experiment, we will observe the show of every group, like cold endurance capacity, provoked degree by snatching, run distance, autonomic activity, genito-function(male), etc. All the rats would be killed to detect organic pathology when model group' s rats showed the symptoms of kidney-yin deficiency. Results:1. experiment I .-Using Thyroxine solution feeding rats, 12. 5mg/kg. bw per day for 21days, rats' Cort, T, E2, T3, T4, cAMP, cGMP, and organic pathology results accordant with pathological changes of Kidney-yin Deficiency. Establish the dosage and time of making Kidney-yin deficiency rat model would be: Thyroxine solution, 12. 5mg/kg. bw, 21days.2. experiment II: Feeding rats with Thyronxine solution 75mg/kg. bw and Reserpine 0.5 mg/kg. bw per day for 10 days, rats' Cort, T, E2, T3, T4, cAMP, cGMP, and organic pathology results accordant with pathological changes of Kidney-yin deficiency. Establish the dosage and time of making Kidney-yin deficiency rat model would be: Thyronxine solution 75mg/kg.bwand Reserpine 0. 5 mg/kg. bw, 10 days.3. experiment III: Making the Kidney-yin deficiency rat model by the way established in experiment I and'experiment II. During the process, rats gradually losing weight, become emaciated, depilation, coat become withered and yellow, and behave some symptom, such as fizz and frightened. The average time of swimming in cold water, the ability of autonomic and burdened activity are lower than blank group, the genito-f unction (male) are greatly decreased too. Liuweidihuang Wan group' s rats have recuperated after treated by Liuweidihuang Wan. The numerical value on T/E2, T4, cAMP/cGMP were improved than model group' s (p<0. 05), T-, and organic pathology results has showed the trend of enhanced. Thus, Liuweidihuang Wan is medicable for Kidney-yin deficiency.4. experiment IV: Liuweidihuang Wan group' s rats have recuperated after treated by Liuweidihuang Wan. The numerical value on average time of swimming in cold water, the ability of autonomic activity and the genito-function were improved than model group' s (p < 0. 05), and organic pathology results has showed the trend of enhanced. Thus, Liuweidihuang Wan is medicable for Kidney-yin deficiency.Conclusion:The preferable method of making Kidney-yin deficiency rat Model with Thyronxine is: SPF male SD rat, weight 200-240g, fed Thyronxine solution 12. 5mg/kg. bw per day for 21days could be used as Kidney-yin Deficiency animal model.The preferable method of making Kidney-yin deficiency rat Model with Thyronxine and Reserpine is: SPF male SD rat, weight 200-240g, fed Thyronxine solution 75mg/kg. bw and Reserpine 0. 5 mg/kg. bw per day for lOdays could be used as Kidney-yin Deficiency animal model.
