Objectives To investigate the effect of C-myc antisense oligo-deoxynucleotide (ASODN) on growth ,apoptosis and cell cycle of HEPG-2 cells.Methods HEPG-2 cells were cultured conventionally. Growth inhibition rate of HEPG-2 cells ,after having been treated with C-myc ASODN, were detected by MTT method. Morphological change of cells were observed under a fluorescent microscope. DNA ladder was examined by DNA eiectrophoresis. The apoptotic rate and cell cycle phase distribution were examined by flow cytometer analysis. The expression of C-myc gene was detected by immunohistochemistry staining and RT-PCR on the mRNA or protein level respectively.Results The result of immunohistochemical and RT-PCR assays showedthat C-myc ASODN could decrease the expression of C-myc gene . The growth inhibition of HEPG-2 cells, after having been treated with C-myc ASODN, was certained by MTT analysis. Furthermore ,with the concentration increasing of C-myc ASODN and time passing, the inhibition ability also elevated. Cell apoptosis was identified by fluorescent staining, DNA eiectrophoresis and flow cytometer. At the same time ,C-myc ASODN could block cells cycle on G0/G1 phase by flow cytometer analysis.Conclusions (1) C-myc ASODN could inhibit the growth of HEPG-2 cells.(2) C-myc ASODN could induce apoptosis of HEPG-2 cells. (3)C-myc ASODN could block cells cycle on G0/G1 phase. |
