Objective: The antivirus research of natural active substance has developed an new antivirus treatment technology in latest years and has displayed a great application potential.indole-2,3-dine(ISA) is an nature ocean antibiotic extracted from lobster-an ocean organism.Now ISA has been confirmed exsiting in natural indigo-a traditional Chinese medicine andhuman body as an endogenous active substance. As it has been reported that some derivative of ISA has antivirus action, but the reseach of activity in ISA antivirus activity has not been reported . The antiviral effect of ISA was investigated by Establishing the BHK-21 cell models and animal models infected with Sendai virus in this paper;Methods: Establishing the BHK-21 cell models infected with Sendai virus which was cultured and separated in chicken embryo, the antiviral effect of ISA was observed by cytopathic effect (CPE) method;The antiviral effect was also observed by different administration time;its effect on immunological function was observed by hemagglutination inhibition test using sendai virus infected mice model;its protection effect to mice which was infected Sendai virus was observed by mice mortality rate and lung index. Results: ISA has showed significant dose-dependent CPE inhibition effect in the anti-Sendai virus experiment by CPE method when the concentration is 39-156μg·ml?(P<0.05), IC50 =37.61μg/ml, TI=11.9;The dose and CPE inhibition rate has significant positive correlation ,r=0.97 (p<0.01). The inhibition effect experiment on different administration time showed that ISA administration on 0,2,4,6 hour has significant CPE inhibition action (P<0. 01);Compared with N.S group, ISA in higher dose (4 mg·0.2mr-1 )can accelerate the production of hemagglutination inhibition antibody in hemagglutination test (p<0.01);Compared with N.S group, ISA in high dose (4mg/0.2ml) and low dose (1 mg·0.2ml-1) can dose-dependently heighten average survival time and decrease mortality rate (P<0. 01);Compared with N.S group, ISA in high dose(4 mg·0.2ml-1 )(P<0. 01 )and low dose (1 mg·0.2ml-1) (P<0.05)can also decrease lung index, lung index=28.7%, 18.2 %. Conclusion: ISA has anti-Sendai virus effect and has a satisfactory margin of safety . It can inhibit SV penetrating the cell membrane, also can inhibit the duplication of SV, but the detailed mechanism is not clear;As hemagglutination test confirmed its enhancement immunologicalfunction, ISA diferred from other drug has more development value . ISA can dose-dependently heighten average survival time and decrease mortality rate and show a satisfactory cell protection;According to Acute and chronicity toxicity test, ISA will be a prospective antivirus drug. This paper provide theoretic and experimental evidence to more investigation for its antivirus effect. |
