Objective To investigate the expression of HIF-1α and TNF related apoptosis inducing ligand receptor in cervical tissue and their correlation with the clinical stage,pathological grade, radiosensitivity and prognosis of cervical cancer. To estimate the state and prognosis of the patient , to study the mechanism of cervical cancer and to find a new way to therapy the carcinoma. Methods The expression of HIF-1αmRNA and TRAIL-DR4 mRNA in 45 cases of cervical cancer were assessed by in situ hybridization histochemistry(ISHH). To analysis their correlation with the clinical stage,pathological grade, radiosensitivity and prognosis of cervical cancer. Rusults1. HIF-1αmRNA was over expressed in cervical cancer 27(60%). In 12 (42.9%) cases of FIGO stage I /II, there was a positive expression, In 15 (88.2%) cases of FIGO stage III /IV, there was a positive expression, The expression of HIF-la elevitated as the clinical stage increased (x~2=9.07, P<0.05) and the pathologic gradedecreased (/2=8.03, P<0.05).The HIF-lamRNA levels in patients with recurrance or uncontrolled was higher than those without recurrance or uncontrolled (^=7.63, P<0.05) . No association between HIF-lamRNA expression and tumor size,histology and radiosensitivity was found.2. TRAIL DR4mRNA was staining in the plasma , the positive rate of DR4mRNA was 57.8%. In 21 (75%) cases of FIGO stage I / II, there was a positive expression, In 5 (29.4%) cases of FIGO stage III /IV, there was a positive expression.The expression of DR4mRNA decreased as the clinical stage increased (#2=9.01, P<0.05 ) .The positive rate of DR4mRNA expression in well-differentiated cervical cancer was significantly higher than that of DR4mRNA in poorly differentiated cervical cancer .(/2=18.18, P<0.05). The DR4mRNA levels in patients with recurrance or uncontrolled was lower than those without recurrance or uncontrolled (^2=11.63, P<0.05) .No association between DR4mRNA expression and tumor size,histology was found.3. No association between HIF-1 a mRNA expression and radiosensitivity.Sixty-seven percent (n =10) of the 15 patients who had an complete response to radiotherapy were found to have positive DR4mRNA expression before commencing radiotherapy, whereas in patients with incomplete response to radiotherapy, DR4mRNA expression was found in only zero.Patients who had anincomplete response to radiotherapy were found to have low DR4mRNA expression, with complete response to radiotherapy. Conclusions1. The expression of HIF-lamRNA in cervical carcinoma elevitated as the clinical stage increased and the pathologic grade decreased.2. The expression of DR4mRNA in cervical cancer decreased as the clinical stage increased . The positive rate of DR4mRNA in well-differentiated cervical cancer was significantly higher than that of DR4mRNA in poorly differentiated cervical cancer .3. The expression of HIF-la and TRAILDR4 can be used to accurately assess the prognosis in cervical carcinomaThe expression of TRAILDR4 was associated with the radiosensitivity in cervical cancer. Further understanding of HIF-laand TRAILDR4 may provide new therapy regimens for cervical cancer. |
