The Expression And Significance Of Caspase-3,8,9 Related To Apoptosis In Neuroblastoma

Neuroblastoma is one of the most common solid tumours in extra-cranial usually found in babies or young children;it account for 8-10% of all pediatric cancers. Cell apoptotic has been characterized by a progressive series of morphological and biochemical changes, it is a mechanism that organisms utilize to eliminate no need cells. Research shows the programmed cell death or apoptosis and its controlling gene abnormality is one of the main cause of tumor mechanism. Caspases protease families are Cysteinyl Aspartate Specific Protease (Casepase) plays a very important role in cancer apoptosis. Laking of the relationship between NB cell apoptosis and clinical prognosis reports. This study has adopted S-P method to detect the expression of Caspse-3,8,9 in neuroblastoma. The aim of this research is to elucidate the possible relationship between NB cell apoptosis and its clinical values.MATERIALS AND METHODS: Fifty-six paraffin embedded specimens including neuroblastoma(NB)25cases, paraganglioma(GN)10cases, ganglio-neuroblastoma (GNB)lOcases were studied and eleven normal adrenal tissues served as control;Immunohistochemistory S-P method was adopted to detect the expression of Caspase-3,8,9 in each group.Results The expression of Caspase-3 in NB was higher than that of GN and normal adrenal tissue (P<0.05);the expression of Caspase-8 in both of GN and normal adrenal tissue was higher than in that of GNB and NB (P<0.01);the expression of Capase-9 in GN was higher than that of NB (P<0.05);Moreover the expression of Caspase-3,8 was related to clinical stage in NB, favorable histologic categories ( I + II +IVs ) was higher than unfavorable histologic categories (III+IV ) P<0.05. The positive relationship was noted between the expression of caspase-8 andcaspase-9 in NB (rs=0.528,P=0.007) ,but no relationship was found between Caspase-3 and Caspase-8, Caspase-3 and Caspase-9.Conculsions Expression of Caspase-3,8>9 can be considered as a useful treatment marker and prognostic for neuroblastoma.
