Objective: To investigate the growth inhibition of antisense oligonucleotides of telomerase on liver tumor cell enhanced by albumin nanoparticles.Methods: Albumin nanoparticles were prepared and solidification temperature , solidification time , stirring speed on preparation of albumin nanoparticles were investigated. There were six groups in the study : (1) blank group;(2) albumin nanoparticles group;(3)free plus sense oligonucleotides group; (4) free antisense oligonucleotides of telomerase group;(5) plus sense oligonucleotides and albumin nanoparticles group ; (6) antisense oligonucleotides of telomerase and albumin nanoparticles group. Influence of antisense oligonucleotides of telomerase on telomerasewas studied by TRAP-ELASA method. The inhibition of antisense oligonucleotides of telomerase on HEPG2 growth was studied by MTT method. There were ten nude mice bearing athymic cancer in every group and injected drugs subcutaneously. 15 days later, weight of tumor was gotten and the growth inhibition of antisense oligonucleotides of telomerase on liver tumor cell was gotten .Results: Albumin nanoparticles could enhance the inhibition of antisense oligonucleotides of telomerase on liver tumor cell and telomerase.Conclusion: Albumin nanoparticle can be used as an effective carrier in antisense technology...
