Objective To validate whether hepatitis B virus deoxyribonucleic acid (HBVDNA) localized in dentitic cells (DCs) in patients with chronic hepatitis B (CHB) and to analyze the relationship between HBVDNA and DCs functional impairment.Methods Mononader was isolated from peripheral blood of patients with chronic hepatitis B infection and healthy controls, further cultured into DCs in medium containing rhGM-CSF and rhIL-4. T phenotypes of DCs were checked by frow cytometry(FCM). HBVDNA in DCs was checked by polymerazl chain reaction(PCR).Results DCs in patients with chromic hepatitis B had lower expression costimulatory molecules and sarface molecules CD1a, HLR-DR) than that in nonnal controls, and especially CD1a exhibited more significant decrease than that in healthy controls PCR revealed that DCs in patients with chronic hepatitis B localized HBVDNA.Conclusion Infection of HBVDNA localizes in DCs in patients with CHB. The expression of phenotypes on DCs in patients with CHB decreases than that in healthy controls.
