Skin hemangiomas is a common and benign tumor, which frequently occurs in infants and children. Excessive proliferation of endothelial cells and vascular rapid growth are the most characteristic pathological changes of hemangiomas. In accordance with endothelial cells growth, mast cell enumeration, fibre cells formation and fatty infiltration and so on, hemangiomas was divided into proliferative phase, involuting phase and involuted phase. Up to date, it isn't very clear of true mechanism of hemangiomas. Recent studies manifested hemangiomas was a benign tumor with vascular excessive formation, excessive proliferation of endothelial cells and angiogenesis were very important for hemangiomas development. Researchers more and more attached importance to study some factors on affecting proliferation of endothelial cells and angiogenesis.Objective: To investigate the expression and significance of cyclin D1 and p27 in the occurrence, development and regression of human dermal hemangiomas.Methods:49 hemangiomas and 5 nomal skin samples around hemangiomas were collected after surgery excising, which all had intact clinical and pathological datum. Hemangiomas was classified as for as Mulliken's classification criterion and the expression of PCNA. We examined the expression of cyclin Dl and p27 in proliferative, involuting human hemangiomas and normal skin tissues by using immunohistoehemical technique. Immunohistochemical technique for factor Ⅷ-related antigen was used to prove that the cells which expressed cyclin Dl and p27 are endothelium. Average light density and positive area rates of cyclin Dl and p27 proteins expression were measured by quantitative image analysis(HPIAS-2000).Results: 1 Phases of hemangiomas The results indicated that there were 27 in proliferative phase and 22 in involuting phase of all 49 hemangiomas.2 cyclin Dl protein expression Positive expression of cyclin Dl protein was located in cytoplasm or nuclei of endothelial cells of hemangiomas, diffused brown-yellow or brown positive particles were observed, less expression of cyclin Dl protein was detected in nomal skin tissue. In proliferative phase, involuting phase hemangiomas and nomal skin tissue, average light density and positive area rates of cyclin Dl protein expression were 0.2903+0.0068, 0.2775±0.0086, 0.2678+0.0011; 0.4448±0. 0918, 0.1409+0. 0934, 0.0862+0.0570 respectively. There were significant differences of cyclin Dl protein positive expression in proliferative phase hemangiomas compared with those of involuting phase hemangiomas and nomal skin tissue respectively(P < 0.05). But no significance of cyclin Dl protein positive expression was observed between involuting phase hemangiomas and nomal skin tissue(P>0. 05).3 p27 protein expression Positive expression of p27 protein was located in cytoplasm or nuclei of endothelial cells of hemangiomas, diffused brown-yellow or brown positive particles were observed, more expression of p27 protein was detected in nomal skin tissue. In proliferative phase, involuting phase hemangiomas and nomal skin tissue, average light density and positive area rates of p27 protein expression were 0.1799±0. 0067, 0.1934+0. 0116, 0. 2289+0. 0164; 0.1589+0. 0736, 0. 3747± 0. 1445,0.8916+0. 1514 respectively. There were significant differences of p27 protein positive expression in proliferative phase hemangiomas compared with those of involuting phase hemangiomas and nomal skin tissue respectively and between involuting phase hemangiomas and nomal skin tissue (P< 0.005).4 the correlation of cyclin Dl and p27 protein expression In hemangiomas, expression of cyclin Dl protein and p27 protein are reverse(P<0. 01).Conclusion: It is suggested that, cyclin Dl might progress the proliferation of human hemangioma endothelial cells and angiogenesis in proliferative hemangiomas; p27 might inhibit the proliferation of human hemangioma endothelial cells and angiogenesis in involuting hemangiomas. |
