Objective: To study intervene effects and its mechanism of grape peel extracts on imitative dementia rats, discuss whether it can improve study ability and better memory obstacle. Method: The 72 health male Wistar rats weighting 180~220g were fed general meal for one week, then divided into 6 groups at random, and every group has 12 rats. According to the document reported, except blank group, other rats were given by intraperitoneal injection of D-galactose 60mg/kg and NaN02 45 mg/kg, Continue 60 days. Simultaneously, administer physiological saline solution,Piracetam Tablets 230mg/kg,low dose grape peel extracts 26. 67 rag/kg, middle dose 53. 33mg/kg, high dose 160mg/kg to intervene respectively. Using neurobehavioral tests , antioxidation genus , cholinesterase genus and nitric oxide synthase, histopathological and morphological parameters and immunohistochemical methods etc indexes to observe and evaluate. Results: Grape peel extracts can significantly ameliorate learning and memory function of imitative dementia rats, and improve SOD, NOS, CAT enzyme activity in rats brain, inhibite MDA, AchE enzyme activity, reduce the expression of P-APP+ and P-AP+ in cerebra cortex and Hippocampus. Conclusions: Direct against "Oxygen free radical damage"and "The choline damage"mechanism , Grape peel extracts can inhibite oxygen fiee radicel to form; ameliorate cholinomimetic neural function; enhance NOS enzyme activity in the Hippocampus and cerebra cortex; reduce the development of p-AP deposited macula; so grape peel extracts probability have a preventive action against early-aged dementia.
