Objective: To study the effect of excessive VitA and Zn intake respectively or unitedly on antioxygenic capability, cell cycle and apoptosis protein in the rat spleen. The study wants to confirm the interaction between VitA and Zn, and provide reasonable foundation for the supplement of VitA and Zn.Methods: Mature rats (8 weeks-old Wistar) were divided into 4 groups. One group (n=16,CG) administered deionize water (0.5ml.d). The second group (n=16, VG) administered retinal acetate (6.27×104IU.kg wt-1.d). The third group (n=16, ZG) administered zinc sulfate (150.49mg.kg wt-1.d). The last group (n=16, UG) administered both (VitA 6.27×104IU.kg wt-1.d and Zn 150.49mgkg wt-1.d). Every group were treated the same mode of lavage. Half the numbers of every group were killed after 2 and 3 weeks. Blood of abdominal aorta and spleen were collected. And then, hemogram examination, serum and spleen T-AOC, MDA, SOD,GSH-Px, CAT and anti O2-., cell cycle and expression of apoptosis protein (Caspase-3, P53, Bcl-2) of the spleen detections were carried out.Results: 1. There were no significant influences on weight, food intake and spleen-body weight ratio of the three test groups compared with the control group. 2. The concentration of VitA in serum and spleen of the UG decreased significantly compared with the VG, and the accumulation of Zn in the spleen was higher than ZG significantly. 3. The total antiocidation capability decreased in spleens of the three test groups which included T-SOD, CuZn-SOD,GSH-Px, CAT and anti O2-., The cell mitosis of spleen in ZG and UG were delayedsignificantly, and there was no significant change in VG 5. The expression of Caspase-3 and P53 protein of spleen in the three test groups were negative, but the higher positive expression of Bcl-2 protein was observed in every test group and their different intensities were ZG>UG>VGConclusion: 1. The toxic effect which excessive supplement this dosage of VitA and Zn in spleen is high than in serum. 2. There is interaction between VitA and Zn in the process of organic metabolism. 3. Excessive supplement of alone or both VitA and Zn enhance oxygen stress and decrease antioxygenic capability in spleen. 4. Excessive supplyment of Vit A, Zn alone or both lead to the over expression of Bcl-2 protein. |
